MoAb Isotype: IgG1, kappa for all MAbs Clones Offered: 3A1, 1G4, 1D6, 1D3, 7D6 Hybridoma clones have been derived from hybridization of X63-Ag8-653 myeloma cells with spleen cells of Balb/c mice immunized with purified human proinsulin. Specificity: MAbs 3A1 and 1G4 are directed to different antigenic determinants. MAb 3A1 reacts with the site of connection of C-peptide and C-end of B-chain of proinsulin molecule. MAbs 1D6 and 1D3 react with different epitopes on human proinsulin molecule; both of each are generated by the interactions between insulin and C-peptide parts of prohormone. Kd for MAb 3A1 is 1.1 x 10-8 M. Kd for MAb 1G4 is 2.6 x 10-7 M. Kd for MAb 1D6 and 1D3 is >3 x 10-8 M. Kd for MAb 7D6 is > 5 x 10-8 M. MAbs 3A1, 1G4, 1D6, 1D3 and 7D6 do not cross-react with human, bovine and porcine insulin, bovine and porcine proinsulin, and human C-peptide. Applications: Detection of human proinsulin in two-site enzyme immunoassay. Recommended pairs for sandwich immunoassay (coating - conjugate): 3A1 - 7F8 (Cat.# 2I1) 1G4 - mixture of D6C4 and D3E7 (Cat.# 2IP10) 1D6 or 1D3 - 7F8, 8E2 (Cat.# 2I1), D6C4 or D3E7 (Cat.# 2IP10) (Could be used for direct coating of prohormone in serum samples) MAbs 3A1 and 1G4 can be used in immunohistochemistry on frozen sections. Specific immunosorbtional removal of proinsulin molecules (with immobilized 1D6/1D3 MAbs) from serum samples before the measurement of insulinic C-peptide.
